TcCYP9E2 Gene Characteristic and Regulating Trehalose Metabolism in Response to High-CO2 Stress

Cytochrome P450 monooxygenase (CYP) is one of the three detoxification metabolic enzymes in insects, and involved metabolism transformation endogenous substances as well activation degradation exogenous compounds. This study aims to reveal molecular mechanism CYP9E2 Tribolium castaneum adapting high-CO2 stress. By predicting sequence function CYP9E2, analyzing temporal spatial expression profile TcCYP9E2, using RNAi silence TcCYP9E2 combined with a stress treatment, we measured carbohydrate content, trehalase activity, gene levels trehalose T. castaneum. A bioinformatics analysis showed that predicted weight protein encoded by 60.15, theoretical isoelectric point 8.63, there no signal peptide, hydrophilic. An evolutionary tree belongs CYP6 family CYP3 group; spatiotemporal results was highly expressed larvae midgut 48 h after injection dsCYP9E2, survival rates decreasing increase CO2 concentration. Under condition 75% CO2, contents glycogen, glucose, ATP, membrane-bound decreased significantly dsCYP9E2. The TRE-1, TRE-2, GP energy pathways downregulated.
